I dont think there is a “most effective” exercise. Personally, practicing your start is probably the most important thing you can do. Get the technique down perfectly and your time will drop tremendously. So I guess it is 10 yard starts. Primetimes, bounding, and ankle stiffness drills help a lot as well.
I like Barry’s program. It is simple and fairly effective. It obviously lacks dynamic correspondence and the deadlift is hard to master with perfect position and proper hip extension. So I would recommend an RDL in stead, while working on core strength and mobility to get into and perform a proper deadlift. Include some primetimes in the warm up, and then alternate a block of Barrys program with a power/speed block with starts, primetimes, bounds, etc…
